Study on Conditions of Ganoderma Protoplasts Fusion

Abstract: The protoplast fusion conditions between Ganoderma lucidum(GL) protoplast and Ganoderma applanatum(GA) protoplast were compared with chemical fusion and electrofusion. the optimum conditions of protoplast in chemical fusion are: 30% PEG (polyethylene glycol), 0.02mol/L CaCl2 and 30℃ for 30min, with the fusion rate 0.4% , whereas the optimum conditions of protoplast in electrofusion are: voltage 40V, frenquency 2MHz, direct pulse voltage 500V, vector 10kV/cm, pulse 25μs, with much higher fusion rate 40%.
