Establishment of Insulin-Resistant 3T3-L1 Adipocyte Cell Model Induced by Insulin

Abstract: 3T3-L1 preadipocytes were differentiated into 3T3-L1 adipocytes with combined 3-isobutyl-1-methylxanthine, dexamethasone and insulin for establishing an insulin-resistant adipocyte cell model. Treatment with 10-7 mmol/L insulin for 48 h was the best condition for induction of insulin resistance as determined by glucose consumption experiments. Insulin resistance could be kept in the developed model for 48 h. High concentration of insulin could successfully induce insulin resistance in 3T3-L1 adipocytes with the advantages of time saving, good repeatability and ease operation. This model will be also available for screening of natural products that improve insulin sensitivity.
