Optimization Study on HPLC Detection Conditions of Apple Polyphenols

Abstract: High performance liquid chromatography(HPLC) was used to detect apple polyphenols,and its basic operation conditions were determined as:C18 column(150×4.6 mm,5 μm),methanol-acetic acid as mobile phase,flow rate 1.00 ml/min,column temperature 40 ℃ and 20 ℃ and wavelength of detector 280 nm and 320 nm. All compositions of apple polyphenols were separated completely under these chromatography conditions. Phenolic compounds were extracted from apples with ethyl acetate. Neutral phenolics were extracted at pH 7.0 and acid phenolics were extracted at pH 2.0. RP-C18 column is suitable to separate apple polyphenols and the mobile phase for the separation of neutral and acid phenolics is identical,the mixture of 4% acetic acid and purified water.
