碱水解及超滤提取金耳菌丝多糖的初步研究

Study on Extraction of the Polysaccharides from Mycelium of Tremella auramtialba by NaOH Hydrolysis and Ultrafiltration

摘要: 本文报道了用NaOH水解金耳菌丝体,用超滤法浓缩水解液提取金耳菌丝体多糖。采用正交试验筛选出碱法水解金耳菌丝体的最适宜条件:碱量及浓度为25倍菌丝体重量的0.75mol/LNaOH,水解温度为40℃,水解时间为4h。水解液经超滤浓缩、乙醇沉淀、干燥得多糖制品。其多糖含量高于54.69%,蛋白质含量低于2.5%。总提取率为4.5%,高于水提法的多糖得率。

Abstract: NaOH hydrolysis and ultrafiltration were applied to extract of polysaccharides from the mycelium of Tremella auramtialba. The orthogonal test showed that the optimal conditions were: NaOH concentration 0.75mol/L, volume of NaOH solvent 30 times as the weight of mycelia, hydrolysis temperature 40℃, and hydrolysis time 4 hours. The extract rate of polysaccharides by NaOH hydrolysis and ultrafilter was 57.7% and total product yield 4.5%, which were both higher than those aqueous extract method.
