苦荞麦抗肿瘤蛋白的分离纯化及结构分析

摘要: 本研究采用硫酸铵分级沉淀、DEAE-Sepharose FF离子交换色谱、Sephadex G-100凝胶过滤色谱和SephacrylS-200凝胶过滤色谱对苦荞麦水溶性蛋白质(TBWSP)逐步分离纯化,并结合细胞实验,筛选出苦荞麦水溶性蛋白质中体外抗肿瘤活性的有效组分TBWSP31。体外抗肿瘤活性实验表明,TBWSP31对人乳腺癌细胞株Bcap37细胞有明显的增殖抑制作用,并且存在时间效应和剂量效应。SDS-PAGE表明,TBWSP31为单一组分,其亚基的相对分子质量为57000。CD分析表明,TBWSP31的α-螺旋的含量为33.9%,β-折叠的含量为22.8%,β-转角的含量为11.3%,无规卷曲的含量为32.0%。

Abstract:  Water soluble protein of tartary buckwheat (TBWSP) was purified by ammonium sulfate fractionation with DEAE- Sepharose FF chromatography and Sephadex G-100 gel filtration. The antitumor effects of the fractions against human mammary cancer cell Bcap37 were studied with MTT assay. The fraction of TBWSP31 showed that higher antitumor activity, and time- and concentration- dependent antitumor effects were observed. SDS-PAGE analysis showed that the molecular weight of TBWSP31 is 57000. CD analysis showed that TBWSP31 has α-helix(33.9%), β-sheet(22.8%), β-turn(11.3%), and random coil (32.0%).
