响应面法优化玉米须黄酮提取工艺

 Optimization of Extraction Process for Flavonoids from Corn Silk by Response Surface Methodology

摘要: 采用响应面法优化玉米须黄酮的提取工艺条件。以乙醇体积分数、提取温度、提取时间和料液比为考察因素,黄酮提取率为响应值,进行四因素三水平Box-Behnken试验设计,获得多元二次回归方程;考察玉米须黄酮体内抗氧化作用。结果表明:最佳提取工艺条件为乙醇体积分数58%、提取温度78℃、提取时间1.7h、料液比1:26(g/mL),此工艺条件黄酮得率为0.41%。玉米须黄酮能够降低血清和肝脏中的丙二醛含量,增加超氧化物歧化酶活力,具有一定的抗氧化能力。

Abstract: This work reports the use of response surface methodology optimize the extraction of flavonoids from corn silk. The extraction yield of flavonoids was investigated with respect to four variables including ethanol concentration, temperature, time and material-to-liquid ratio. A multivariable quadratic regression equation was built using a four-variable, three-level Box-Behnken experimental design. The optimal conditions for extracting flavonoids from corn silk were determined as 1.7 h extraction with 58% ethanol at a material-to-liquid ratio of 1:26 (g/mL) and 78 ℃. Under these conditions, the extraction efficiency of flavonoids was 0.41%. Oral ingestion of the flavonoids extracted from corn silk resulted in reduced MDA content in the serum and liver of mice and enhanced SOD activity, suggesting that these compounds have antioxidant effects in mice.
