大孔吸附树脂分离富集大麦多酚的研究

摘要: 通过静态吸附量和解吸率的测定对六种大孔吸附树脂进行筛选,然后确定大麦多酚分离富集的最适操作条件。结果表明,AB-8大孔树脂具有好的吸附性能和解吸效果。最适吸附条件为:上柱料液浓度1.0mg/ml,pH6.0,上柱速率为2~3BV/h。最适洗脱条件为:乙醇浓度70%,洗脱速率2BV/h,洗脱体积3.0BV。经过分离富集后大麦多酚的含量增加了3倍,还原能力和清除DPPH自由基的能力也随之大大提高。

Abstract: Static adsorption capacity and static desorption rate were assayed to screen the proper macroporous resin for barley polyphenols, and most suitable manipulation parameters of macroporous resin were confirmed. The results showed that AB-8 macroporous resin has strong adsorption capacity and easy desorption property among the six resins used in the experiment. The most appropriate adsorption conditions are: sample concentration 1.0 mg/ml, pH value 6.0 and sample flow velocity 2~ 3 BV/h. The most fitting elution conditions are: 70% ethanol concentration, elution rate 2 BV/h and elution volume 3.0 BV. After the isolation and enrichment of AB-8, the contents of polyphenols increase 3 times and the reducing power and DPPH radical inhibition are improved obviously.
