Abstract: Corn pollen was processed by different wall-breaking methods before extraction. The orthogonal test design was used in extraction optimization of pollen polysaccharide of corn (PPC). It showed that the pollen wall-breaking process was the mainf actori nfluencing PPCe xtraction,f ollowed byt hep roportiono fp ollent os olution, thee xtractiont emperaturea nde xtraction time. The enzyme wall-breaking method and machine wall-breaking method were better significantly than temperature-differ- ence wall-breaking method, but there showed no significant differences between the enzyme wall-breaking and machine wall- breaking method. As machine wall-breaking had advantages such as convenient and low cost over enzyme wall-breaking method, it was suitable in PPC extraction. For machine wall-breaking method the optimized conditions for PPC were: 1:6 proportion of pollen to solution and 70℃ of extraction temperature for 2 hours.
