In Vitro Assay on Anti-proliferation Effects of Soybean Isoflavones on Rat Breast Carcinoma Cell

Abstract: To study the mechanism of growth inhibition of rat breast carcinoma cells by soybean isoflavones, cancer cell line was treated invitro with daidzein and genistein. Microculture tetrazolium(MTT)assay, 3H-TdR incorporation, and cell cycle flow cytometry were usedto study the inhibitory effects of daidzein and genistein on cell proliferation. Results showed that daidzein and genistein could result in adose-dependent and time-dependent growth inhibition and decrease 3H-TdR incorporation in DNA of rat breast carcinoma cell. Flowcytometry showed that cells were accumulating in the S phase, but cell apoptosis was not induced.
